Splinter Cell Pandora Tommorow online multiplayer. There has been nothing funnier than torturing mercs on the hospital level.
Equally, Midnight Club 3 online racing with power ups. Again, so hilarious icing an opponent at a 90 degree turning making them go straight threw it into a shopping mall or over a bridge onto the freeway putting them out for the rest of the race.
I've never laughed so much in a game as MC3 online.
GTA. Their spoofs of rock music and musicians, televangelists, gangsters, American's gun culture (Ammunation's commercials were awesome),music radio(we play the 13 most popular songs, the only songs worth listening to), talk shows and lots of other stuff I am forgetting are all hilarious.
Escape from Monkey Island. Still my favourite, followed by the humorous execution of Portal.
I'd also like to mention The Witcher, since it has some pretty damn funny moments. Because of the rules, I can't write the exact sentence that ade me laugh, only that it involves dwarf genitalia, and a mother.
I dunno if it was the funniest, but the Bard's Tale would at least deserves an award.
All the rambling against the narrator, yeah, the maincharacter and the narrator are **** each other. But that is merely part of the fun...the evil summoning from whatever deep layer in Hell was also quite humoristic...as was pretty much all the game. :P

Vampire: The Masquerade - Bloodlines. It may not have the most humour but it has the best humour. Some of the off-colour jokes just make me laugh out loud for quite some time. I know last night I was talking with Smiling Jack in the Last Round and just couldn't help but laugh at something he said at a completely random point in the conversation... which oddly enough, I don't remember.
A lot of the anarchs are hilarious as are some of the Kine who roam the streets.
Portal was pretty funny despite my doubts, team fortress 2 is absolutely a riot. Halo 3 had some very funny lines in it. Paper Mario 1000 year door made me laugh a few times.
hmm phoenix wright has made me laugh quite a bit, and ff5 has it's funny moments as does ff7. Portal sounds hilarious just a shame i don't have the console for it :(
Funniest game ever is Conkers Bad Fur Day hands down, and Banjo Tooie is pretty damn funny too, mainly because of kazooie's smart remarks about everything.
