I just bought my first Wii. Is the disc slot supposed to be glowing blow constantly when it's turned off...?

Yea thats a default setting. You can turn it off by selecting no for slot illumination under the wii system settings.

It signals that you either have an unread letter in the message menu, or that there is a system update available. You can turn the disc illumination off if you want, it isn't really important :P

its supposed to do that when its on i like it blue but its on default settings which can be switched on or off at any given time.

It will NOT glow blue all the time; only when you receive a message to your wii message board.
