its different for every game that supports it. I only have done it on Metroid Hunters. You can talk before and after matches by holding a button (forgot which one).
Keep in mind that Nintendo doesnt know what they are doing with online, they are practically playing it by ear......
Well, what i do, if i get whooped or something, i cuss out my opponents after wards. But my DS dont have a microphone thing so only i can hear myself cuss them out

Depends, Super Princess Peach uses it when you're in the submarine underwater. You have to blow in the mic then in the game, bubles will come out of the submarine and whatever enemy it hits will get hurt and possibly die.
Another one I know of is Trace Memory, various puzzles require you to blow into the mic to blow off dust off objects, or blow out candles to get a better view of things.

I think for Metroid Prime: Hunters, you need your opponent's friend code to talk with them. You might know this, but you can also type what you are saying to your opponent in mph also.
