For me, I`ve invested the most time altogether in luminous arc.

Depends if you want single or multiplayer. Mario Party for example gets old pretty fast with single player, just because it's more fun to play with other people. But anyways, I'll list a game with good replayability in general:

-Mario Kart (as mentioned)
-Bomberman Land Touch
-Advance Wars: Dual Strike
-Meteos
-Puzzle Quest
-Nintendogs
-Elite Beat Agents

Also, FFIII is a relatively long RPG game and I've heard that Lunar Knights is also a lengthy game.

Pokemon for most of people, but to me its too slow.

Advance wars was great, maybe litle too short.

Castlevanias are good..

And megamans.

And brain training games maybe.

Pokemon Diamond/Pearl because of long-term playability, same with Tetris DS, MarioKart DS, and Animal Cross Wild World. There are a few others, but my brain is not letting me think at the moment.

Besides what's already mentioned...

Obviously Super Mario 64 DS and New Super Mario Brothers. Those games are pretty much must-owns for DS owners.

And also I have to say Sonic Rush, because it's one of the few good Sonic games recently that stands out for itself: being fast, 2D, kickass boss battles, and it feels like the 2D Sonic games in some way...except it's not quite as good.
