That depends on whether or not you want the 20-40 hours to be part of a cohesive story...or just simply are looking for that many hours of gameplay. From a gameplay perspective Advance Wars Dual Strike can keep you busy for a long time (although it lacks WiFi, but its sequel coming out is going to have it). If you want it to be a part of a single player campaign then go for Final Fantasy III or pre-order the re-make of IV coming out to the DS. You will be able to get a lot of mileage out of any of those titles.

Since GBA games are playable on the DS you could do leaf green. I'm currently at almost 48 hours. Or any pokemon game except R/B/Y/G/S/C. They all are well made and time consuming.

Puzzle Quest hands down. It doesn't keep a running timer, but I know I have clocked well over 30 hours. If you like bejewled and RPGs give it a try :) I love it!

Puzzle Quest or the newest of the Harvest Moon Series - Rune Factory. Just got it yesterday and I know I will be playing it for many days to come.

Super Mario DS really takes that long to beat? Anywho I played JUS and Pokemon Diamond for a very long time. JUS also has fantastic replay value.
