Some of this is my own wishes and some of it is more or less confirmed. Needless to say, the 3DS should have everything that the DSi has but with the following differences.
Two identical 3D(duh!) widescreens with 1280x768 resolution, mulit-touch and 1024 level pressure sensitivity. (Enough hardware to play good quality 720p movies wouldn't hurt either.)
Analog control stick + d-pad and regular buttons.
If there's no GBA slot I would like the 3DS games on cartridges that are a bit bigger than NDS because I like the fact that developers have room to add to the hardware through the cartridge, that's one of the main reasons why cartridges own, it does in fact prolong the life of a console when the hardware development can follow the games and not just every 5 years or so.
Built in 3G modem and support for IEEE 802.11b/g/n with WPA2. 3G modem would increase the versatility of the device tremendously as you could be online "anywhere", even more important is the support for WPA2, WEP which is the only encryption that DS has is a joke and as a result I'm not using the wireless function on the DS. With WPA2 and a strong key you're much better off.
Advanced tactile feedback, buttons that could push back for instance could probably be used for something creative.
Built in LED projector with 1280x768 resolution and a 2-3 meter range, best used with external battery pack or plugged to the wall.
Wireless support for the Wii controller(s), put down the 3DS, turn on its projector and you and a friend can use a pair of Wii controls to play games, z axes with the remote will act as tapping with the stylus.
Media device/browser functions of the 3DS would be linux based and completely open for modification. For instance you could install the Ubuntu 3DS dist and use VLC to play movies through your WLAN and output it on the 3DS projector.
A device like that would sell better than hotcakes. Think about it.
