I play it on the third (second highest brightness level setting). I find very little difference in light luminosity between the second last and final brightness setting. And at the same time, I reduce the amount of charge cycles the Nintendo DS will have to go through, without having to resort to dropping brightness to 2 (a bit dark) or even 1 (wayy too dark, you're wasting away the most important aspect of the DSLite over the DS Phat, the LIGHT! XD)
This hand of mine glows with an awesome power. It's burning grip tells me to defeat you!!! Now here I go, SHINING FINGER!!!!!!!  But on a serious note, second highest or lowest, depending on battery life. Second highest for not wasting battery too fast while providing enough light. Lowest should be obvious enough.
