Does it matter? None of us will be alive to confirm or care. What matters is that he is here now and he is better than ever.
I am concerned whether a hundred years from now, the franchise is relevant and alive as it is now. I will not know about that as I would no longer be alive to see it.
Mickey Mouse was created in 1928. He's not too far off from his 100th b-day, and I'd equate Mario's popularity up around his area. I wouldn't doubt it, if Nintendo can keep finding ways to keep him relevant.
I think a lot of that depends on what Nintendo is able to do in the coming years on a larger front, honestly. Just yesterday, I was talking with some members of my family about Christmas stuff, and when I told them I still liked Mario-related items, their response was, "They still make Mario stuff?" This kind of question seems to come around every time Nintendo's platforms start to lose relevance; I certainly didn't hear these kinds of remarks during the high point of the DS/Wii days, that's for sure. Yeah, I know it's just one person and a few of his family members, but I feel Mario's relevance will very much depend on the relevance of the company's future systems. I think we're starting to get to the point where Nintendo's games alone don't always sell themselves, and if that's the case, Nintendo has to do something to reinvigorate the company's brand moving forward in a very big way.
When gaming takes it's next step (like our step from 2D to 3D), if Nintendo can make him relevent again and fun, then its a yes, yes and yes.
Simply put...no. It will die off just like Call of Duty and Assassins Creed in a few years. I-for-one have been playing Mario for the SNES and N64 because those were the true Mario games.

One hundred years is too much of a stretch to predict, but what makes you think it'll die off in a FEW years??
