I'm a devoted AC fan. After the semi-disaster of City Folk, I think Nintendo finally understands we need a refresh. The screenshots look good so far. I'll be buying it no matter what, simply because I have every AC game.

Yeah this is the game I'm looking forward to most, with Mario Kart second. I've only been playing AC since Wild World, but having sold that when I got City Folk I really miss the portable element. I've still probably sunk about 70-80 hours into City Folk, but I dread to think how much time I spent on Wild World as even if you're watching TV you can still be fishing or collecting fossils etc.
I want the City to disappear (having to get the bus to that crappy little square just to buy the dodgy paintings etc is a hassle) and I want new stuff to do, but one way or another I'll be buying it, as for me, AC is about 10 times better when it's on a handheld. The pointer on the Wii version is a hassle for me as well as I have to sit close to the TV, pointing the remote away from the screen while walking around. Wonder if you can do the import with City Folk to get items for the 3DS version, like you could between WW and CF. Be nice if I could keep all the Nintendo themed stuff I've been collecting.
I loved AC on the DS. Completed everything EXCEPT for getting Nook's store improvements. Made a friend online, invited him over - and he destroyed my entire village with seeds. So if you can complete everything WITHOUT inviting a 'friend' over, I'd be willing to get it.
No. None of them have been enough of an improvement over the GCN one to warrant buying. Though it's kind of hard to seem like a new game when anytime you start a new game it's completely randomized.
Whenever I get around to buying a 3DS, AC is pretty much a must. The graphical improvements and changes alone are a good selling-point, but if the game has new features or utilizes the 3DS's hardware in some way then it will be a proper sequel.
Based on the early screenshots and information, yes, there's enough changes/additions that it's likely worth the purchase.
