Pain of loss is worse as you know what you are missing. Pain of nothingness can be bad, but is a consistent kind of background pain.

Anyway TC, to answer your question: to have something and then lose it is more painful. Otherwise you're simply living in ignorance which isn't painful at all, just emptiness.
