Warhammer 40K: Space Marine sounds like what you are looking for. I find the single-player PVE campaign on NORMAL to be in many cases harder than the player-vs-player if you can believe that.
The game is very action-oriented and challenging, made so by the health regen system. You have to kill an opponent while they are stunned to regenerate health. (There is no other way.) But what makes it challenging is that different enemies get stunned in different ways and some of them can't be stunned at all. They just die when you impact them, ironically leaving you sitting there like a sitting duck with a smidge of health.
The upside is once I bother to look up how to stun them all I imagine a 2nd playthrough might be looking pretty good. Oh, and the player-vs-player campaign is an absolute blast. Oh, and you're taking part in a lore/mythology/universe that's been around since X (or tabletop RPGs.)

I'm playing this now and you are correct, it is a blast and can be very challenging. I'm on my 1st play through on Normal Difficulty and it can be quite tough in some spots. If you enjoy the Warhammer Universe, which I do very much, , it's a great game and I highly recommend it.
Another suggestion would be the original FarCry, was and probably still is a hard game to get through...also quite long.

I'd feel really stupid right now if I didn't predict above that I was doing something very wrong to be making the normal setting soooo hard. Here it is:
The critters that are so low level they explode when you hit your stun move CAN be used to increase heath. You just have to go straight for the execution move. [/facepalm]
The devs make going back to previous levels really easy in this game which I've already begun. I don't think I'll forward back to the end. (Did I just say that?) I'll just stay right back here and experience the game as it was meant to be. I'm pretty much used to being always backlogged on games any way.
But WOAH you really do feel like your sloggin' through a battlefield. Enemies are just obstacles to convert to carnage along the way. Now, I know how it feels to be a boardpiece in front of those warhammer nerds at Lakewood Mall in 2008. Who knew?
