Because pieces of installed and uninstalled files get left behind over time and clutter the HDD. Defragmenting can help alleviate this but still as Windows gets older and older it gets more and more cluttered and as a result gets slower. Also more background applications might be running in the background and being told to run at startup. This means it takes longer for Windows to boot and they are using up more of the PC's memory.
Just scrubb your pc from time to time by digging around and deleting old empty folders and shtuff. I defragment and have recently tried pc clean up software and so far so good. But honestly, the windows cleanup utility seems to do a fine job.

Folders wont slow down a PC, its caused by background processes that are starting automatically with Windows Statup. To make it faster run MSCONFIG and turn every non-Microsoft item off apart from your AV.

Still a good idea to keep your disks defragmented, plenty of free disk space, and don't keep programs installed that you never use, clean the regisry once in a while. If you do these things, windows stays running pretty well compared to when you first install it.
first of all your pc hdd gets fragmented and you need to defragment it regularly. same happens with ur registry, so try finding a software that can defragmnt both ur registry and ur hdd.i will reccomend tune up utilities 2010 for this. head over to this site and download the trial... http://www.tune-up.com/download/

um, in practise no. When my drive is fragmented I notice a huge difference. Whether I use a third party defrag or windows defrag I get the same results, only difference is the windows defrag takes twice as long. And the defrag can be set to be done on a schedule, it doesnt just run all the time (third party or windows).

I am gonna have to agree with Daytona, useless processes that use up cpu usage will be the number one thing that slows your computer down. Theres no need for a third party defragmenter, just use the one under system tools, it isn't like it doesn't work as well.
as mentioned do a start>run>msconfig and get rid of the **** software puts on your startup. It's quite shocking what gets put in there and how useless 90% of the stuff is. Like why do I need a quicktime service running constantly? And if you have any autodesk products you should see what they put in there without your permission...
