It depends. In FPS, the first thing do when I first play the game, I just go nuts and run at the enemies.

depends what genre... for RPG's I immediatly focus on combat and raise my characters a little bit before starting any main quests/story, also finding out what (if any) combos are effective, spells, weapons, etc.
for shooters I just test out the weapons and mechanics to see what I can do.

Shooters: Find out what the hell the controls are - so many different ones out there.
RPG's: Start the main story.
Fighting Games: Well...uhm, I'll let you answer / figure that out.
Action / Adventure: Uhm...not really much to do except play the main game.
Sports: Read "Fighting Games"
Rockstar Games: Beat the **** out of everything I can.
Puzzle Games: Once again, read "Fighting Games"
Explore everything in the area around me from top to bottom, get used to the controls, basically the basics.gohantech00
yeah i do that for RPGS
for FPS i just do training level if war game if its not I just mess around or I try kill myself in as ways as possible or kill my teammates

Same thing here. I also skip cutscenes first. I start playing seriously the next day.
First mess with the controlls to know wich button is wut... then play the game for hours until realize im tired
