Even though I mastered the fight years ago, Alma from NGB is always the first that comes to mind when I see this question. That bitch probably killed me 50 times before I managed to barely scrape by.
Mine was that chick Jeanne from Bayonetta. It took me about 10 tries to beat her on top that dang rock.

Oh, in recent times, this. Father Gascogine was at least a dozen fights for me, probably more.
Vicar was a one off I'm pretty sure though. :P

I had to just swallow my pride and call help on both these bosses lol. And I think Blood Starved Beast was another one I needed help with.
It was weird, this was before the game had 'clicked' for me (once it did, I think right after Vicar Amelia), I got through the game, not with ease, but with nothing ever being quite out of my reach. These two bosses though, were basically me at my most hopeless.

i dont know why but i always lose to the first boss in skyward sword.....then in the rematch i kick his backside. i have some sort of mental block there :S.
there is a boss in prime 2 echoes where the whole battle is in ball mode and he is quite frustrating.
oh and barrioth in monster hunter 3 (he was made easier in ultimate i think). that is one tricky monster.

Laurence the first vicar, now that is a hard bosses that almost made my summon help for the first time ever in a souls game. I persevered though and managed to get him on my own. Most difficult bloodborne boss and my ass was destroyed over 20 times.
Blood starved beast is so easy. Don't lock on and roll on his sides and try to counter him with your gun. I recommend attacking him then at the last bit of his health counter and punch a hole to finish him off.
Vicar Amelia was so easy that it felt like a dance lol. I love these games
Ebrietas was insanely difficult. I don't dare to fight her again. Thank god she is optional.
@charizard1605: it gets easier if you learn how to control your fast step and go in at the right moment. The animation and attack pattern reveals their weak areas.
Laurence is another story. 2nd phase is incredibly hard to get a chance to strike. Cheap shit tbh.

lol it's weird man sometimes I just randomly go into beast mode. It's like those reflexes from the 8-bit days just kick in and I'm fighting as if my life is on the line.
I also remember a lot of people talking up a lot of shit about The Four Kings fight in Dark Souls. I was dreading the fight man. Then I got there and I think I got hit like once lol.
But yeah man, Father Gascogine... I think most people have trouble on that one, he's somewhat a rite of passage. It's like you're going to learn this game right now or it's time to quit lol.

Man, maybe you wiped the Father out easily, but I know a lot of people had a time with that one. He was definitely the toughest boss in the game for me, aside from maybe fat spider caterpillar thing (I'm too lazy to look it up).
@ConanTheStoner: Yeah, Gascogine really basically is a test of the basic skills required to play the game- I mean, quick reaction times and reflexes, sure, but also staggering, proper use of firearms, knowing when to heal and use items, and so on. He gave me no trouble on my second playthrough- but that first one made me miserable haha.

I mean, I know how the bosses are defeated, the trouble is it was hard for me to pull off the first time I was playing the game, it's why I had problems with them lol.
@ConanTheStoner: Father Gascoigne was basically the test for players to see if they can bear to defeat him. He teaches you how to get used to fast enemies and how to control your step and counter attacking. The first time I fought him I was already dead type of situation but somehow I beat him. The second and third times were a cakewalk because I just know when to attack and counter. I even wore his outfit because he's so awesome lol
@charizard1605: I remember reading that you said Bloodborne is your first souls game so totally understandable for a new souls player.
The animations in bloodborne are faster too so I had to adjust to it too. Not to mention it's an offense type of game unlike the previous games where you can tank and beat them. After bloodborne I stopped using shields in souls. It's much more fun that way.

Scholar of the First Sin was a bitch regarding that.
This one recently. I finished Intrusion 2 last week. Easily the highlight has to be some of the most inventive and satisfying bosses I've seen in years. One of the greatest platforming shooters made by one guy , with wonderfully difficult bosses and constant enemies which require different strategies to defeat, as well as one of the best final bosses in any game ever.

How about Ludwig? So called hardest mandatory boss in the DLC and bloodborne?
I kept hearing how hard that was and I died three times to learn his attacks and all that and then I walked in and kept staggering him and destroyed his ass. The two sorceress after him were more difficult lol.
I honestly think that Laurence 2nd phase is the hardest boss in the game. The orphan of kos was pretty awesome too and his second phase is chaotic and so fast with all the lightning Storm going on that he creates.

I'm not good at remembering boss's names but I know a few on Bayonetta gave me a hard time. Also a few through the FF series One I remember in particular was that mushroom thing toward the beginning of FFXIII, when it's just Lightning and Hope. And the eidolons.
Darklurker is an asshole. Beat them once in my first playthrough but can't in the second, wish you didn't have to give up a humanity each time you try.
Oh and Genshin in Ninja Gaiden II...

If you have not played wings of vi, you do not know what a hard boss is, every boss in the game(except the first 2) are incredibly hard and it culminates into what is hands down the hardest boss I have ever played in my life. Took me a little about 450 tries to beat this douche but watching him die was probably the most satisfying experience in my entire gaming life.

Savage Deviljho kicked my ass for a while. But he turned into a pretty nice suit eventually.
The neutral boss from SMT Strange Journey. Though it is possible to cheese her by building an all reflective party.
