Yeah sure, I do it all the time. But I'd suggest installing the game to your hard-drive if you're goin to play it for long periods of time. Better for the disc.
I leave discs in there all the time and have never had an issue. I've also moved the system, while turned off, with a disc in the tray and haven't had any issues.
Just dont move the console with the game disc inside otherwise it will be fine.

You do realize as long as its off the disc wont mess up? But either way its fine. There's been reports of whether it's off or on and you move your console it may harm the disc.
