Personally, I'd go with Mass Effect.
Assassin's Creed, although enjoyable, does not live up to the hype. I could go into details on this one, but EVEN if it did, it still wouldn't be worth buying over the other games because of how short it is in comparison. So I would suggest renting it.
As for COD 4, fantastic but short single player campaign, and the best multiplayer experience on the 360 (aside from maybe Halo 3, depending on who you ask).
As for Mass Effect, I figure you already know all about it.
If you love online shooters and are the kind that plays them a lot, you might be able to squeeze more life out of COD 4, but I would still recommend Mass Effect over it, due to my own personal bias. Please note, the only game of the three that I have actually played is Assassin's Creed.

How in the world did it take you 40 hours??? I just killed the 5th dude and I've put in 10 hours max. Judging by the fact that there are nine in total, I am more than halfway done. Most of the reviews I've read said 15-20 or so hours as well. Of course, I am not an achievement fanatic so I could really care less about the flags, and I don't go out of my way to do the templar killing mission either.
