GHWT is definitely more challenging so if you like hard rhythm games go with that. However RB2 has a great set list and the downloadable tracks are amazing.
Lets get this quote straight. GH:WT is "more challenging" because Redoctane/Neversoft/Activision overcharted nearly all their songs. They incorporated unnesceary chords and notes where they shouldn't be just to make the game "more challenging". Now, if you want more realism go with Rock Band. Rock Band has a much cleaner layout to their interface, and a wider song selection (via DLC). Guitar Hero is still stuck in that "Tony Hawk" era with it's layout. About the only thing GH has over RB at this point is their guitars are a lot better. Guitar Hero will continue to sell though because it's got the name that sells and it still appeals to the general population who doesn't really know any difference between the two games. Be smart and research...Rock Band is much much better.
I say get Guitar Hero II or III with a guitar, and play that for a bit. Then when had your fill of that, get the Rock Band games alone and use your Guitar Hero Guitar. I go with Rock Band.
Before GH:WT, it would definitely be Rock Band. Now though since they have the same concept for downloadable songs it all depends on the type of music you like to listen to. Guitar Hero recently signed exclusive rights to Hendrix and I think also The Who. Rock Band has AC/DC and The Beatles, so just look at the tracks available for each and make a decision based on that.

I agree Guitar Hero has so much false overcharting its not even funny.
