Based on the pattern Rockstar developed with the previous gen of consoles (releasing GTA III and then two "spin-offs", Vice City and San Andreas, and then releasing spin-offs of first GTA on that gen and the first spin-off, Liberty City Stories and Vice City Stories), I would say that the next GTA will be based in San Andreas if it's on the PSP, and Vice City/related if released on the 360/PS3. Or it could be different if Rockstar decides to switch things up.
Personally I couldn't care less. GTA games are always of high quality regardless of the area they are based in; all I'm concerned about is the gameplay, which Rockstar did an amazing job of enhancing with GTA IV.

Well i'm guessing it will probably take place once again in Liberty City. Not that I want it to take place there though. I really enjoyed the location(s) of San Andreas because of it's variety and more rural and suburban feel. It had different areas, some took you through mountains filled with trees and trailers all the way to the plains, with plenty of in between suburban areas. It gives a great feel of having to travel to great lengths and noticing the scenery rather than once you get to the next island thinking "Haven't I seen all of these sky scrapers and city lights in the last island?"
If you want my personal opinion on it though, I'd say it should be set on an area in Europe. It will probably never happen seeing as the game is set in the United States, but I think it'd not only be a nice change but a great setting. The possiblities are endless and the fact that Europe civilizations have been around longer could give the game some character it hasn't had before. Imagine that you're running from the cops in Venice, Italy. Because the city has waterways rather than streets, the chase could be very interesting, jumping from a rooftop into the water, swimming away through the maze of boats. Or imagine that you're on your last leg, about to be cought by a rival gang/mafia, etc, and your find your self at an old European castle from the Dark Ages. You could still have some larger cities areas of course, but to throw in a little old century charm would be pretty cool.
Man it will be 2010 before we see a full new game for even a preview so they better make it big for me to be intrested. I would say the whole fing world. I should beable to take plane flights to newyork to seattle, then fly to Mexico for some drug runs then off to russian to vist the KBG. Then go visit some girls down in brazil and so on. New generation new gameplay right?
They better make it big or I'm not intrested I didnt' like IV so they better do something to gain my intrest back in this game.

Yeah. I thought that Liberty City would be bigger than San Andreas in GTA IV, but now that I realize that it isn't, playing at San Andreas would make things so much cooler. Long drives ftw!
You guys are thinking to basic when you say Tokyo. Thinking about it more deeply the geography of Tokyo is terrible the populous per km of land is more than the land itself. The roads and streets would be far to congested making speeding almost impossible before hitting something, I'm guess less than 50 feet at max. Don't try to pull Tokyo drift out your ass because that was the worst "Fast and Furious movie ever". I want to see the San Andreas setting again.
