There were a lot of stellar games this generation, so it's hard to choose one.
I'd have to nominate RDR, L4D2, Dark Souls, Skyrim, Oblivion, Borderlands and CoD4, though (several games beyond those, but I only wanted to put games I'd actually played on 360 as opposed to PC).
But that would leave out Bioshock, and I wouldn't want to do that.
As the one poster pointed out, they all do kind of different things, so it's hard to choose.
If I HAD to I'd probably go with Dark Souls, with RDR running a close second.
This would just turn into people's favorites - mine with fair criticism:
Borderlands - mediocre story
Dead Space 2 - personally found nothing wrong with it (too short?)
Bioshock - too easy even on hard mode
Metro 2033 - too niche according to popular opinion
Batman AA - kinda short but for what it is it's near perfect
Singularity - shooters will never be GOATs
Tales of Vesperia - JRPG will never be GOAT in USA
FF 13 - admittedly slow to start and off-putting to new gamers but great strategy element
Mass Effect 2 - thought they made it too simple but story is still great
NFS Hot Pursuit - best racing game (non-simulation of course) I've ever played
This new game «Asura's Wrath» is really making me crazy. I already played the demo 3-4 times since it came out, I'm crazy for that game.
"Masterpiece" would imply that the game is of the highest quality across all possible areas. I would most likely go with something like Gears of War 3. Great single player campaign, phenomenal multi player, and unrivaled co-op. It is, without a doubt, one of the 2 or 3 most complete titles to have been released this generation. But then you have games like Skyrim which don't have Co-Op or Multi player, but have unrivaled single player options. It's just to difficult to choose just one. ACx7This too. The amount of content and value for the money Gears 3 gives is astounding. No matter what kind of mode you like in shooters, Gears 3 has it.
This generation is not over yet.
That being said, it is still very difficult to pick one because different games exist to do different things. Forced to pick I'd say Bioshock.

This, mainly because it improved upon Halo 3 and it's one of the only games I've continued to play since release.

Assassins Creed (all of them, even the first one which I love).
Alan Wake
Borderlands
Bayonetta
Splatterhouse
Skyrim / Oblivion
Mortal Kombat
Dark Souls
Batman AA
Red Read Redemption
Bioshock 1
Lost Odissey
Dead Rising 1
I think they all did extremely well the things they meant to do.
BTW I'm leaving a couple of games out but I don't have my collection near menow :P
It's not my favourite game this generation, but I very much consider Bioshock to be a masterpiece. The concepts, the setting and the script are just sensationally woven to make a game.

Individuals such as Michaelangelo & DaVinci spent at least 30 years of their lives honing their skills before producing their "masterpieces".
There are lots of very good videogames out there, but i still think they all fall short of this elusive status.

Seriously dude...For this I'd have to say Metal Gear Solid 4. It is the perfect example of what a video game, and a sequel should be (the other MGS games are as well, but I'm speaking specifically about this generation).
The story was top-notch (although complex) throughout the whole series, and the telling of the story was executed to perfection. It concluded the saga of Solid Snake in an epic manner.
The graphics (for its time) were amazing, and whats most impressive is that it was all done in real-time, including the cut-scenes. For anyone who played this game, the epic split-screen Snake vs.the Geckos/Raiden vs. Vamp battle comes to mind when thinking about this. Also there were very very few graphical glitches which is important.
The gameplay improved upon its predecessors in countless ways. The controls were completely revamped and alot less frustrating than the previous ones. You could literally play the game like a stealth action game and as a third-person shooter at the same time. The boss battles were very diverse and challenging, not to mention the beauty and the beast corps ladies were gorgeous. The Big Momma set piece and Metal Gear Rex set-piece was epic as well.
The voice acting was superb, the soundtrack and musical score add a ton to the overall gameplay experience too although Im not very knowledgeable about this aspect.
This game incorporated these things, and a lot more, and did so in a fashion that very few games (and I mean maybe 3 or 4 other games ever) have been able to do so well. Anyone who played this game all the way through would be able to vouch for me.
