I say, the final boss in Contra for NES. I was just starting out as a gamer. Young, immature. A horrible noob. By the time I finally made it near the end. I was almost out of life, each time. Didn't understand basic gameplay tactics. It was a major struggle for a whole summer. (Fun struggle)
But defeating the game of Contra. Was also, by far one of my greatest gaming moments. Took me most of the summer, back in the 80's. But when I finally downed it. I ran up and down my street, yelling. "I BEAT IT!"
Funny to think about it now. What others might have thought, I was saying. lol!!!:P

so true, it took me about 12 times to beat him,
about ffxii last boss, he was quite easy for me, barely healed the party, probably because i lost my way at some part of the game and leveled vaan to lvl 79 and ashe to 68...
I found out about a boss on Final Fantasy XI that hasen't been beaten at all, even after a year of the game being out.
God of War on God mode trying to protect your family. Its hard for the first 5000 times you try but you get the hang of it.
Also Any boss from Devil May Cry 3 that game is sick i love the intensity.
