Really? Just from E3:
Mirror's Edge - a unique game in which you're discouraged to fire a gun that focuses on building up momentum to perform parkour moves and features a complete lack of HUD
Dead Space - a unique game featuring mutants that grow new limbs and other body parts if you injure them, which travel on all four walls such that you can easily lose sight of where they're attacking you from and is set in outer space such that you can float around and push yourself from one wall to the other so you're not limited to walking on "the floor".
LittleBigPlanet - an original game featuring "coopetition" with your "frenemies", allowing you to build your own obstacle courses and featuring a unique art-style and unlikely hero known as Sack Boy. Hell, this game can even be used in lieu of a powerpoint presentation.
Fallout 3: The Fallout series has a unique combination of RPG and shooter elements and counts among the few games that pull that off successfully. The VATS system shown off blends traditional shoot-their-weak point with dice-rolling chance-of-success RPG gameplay. And the Fallout series has always been original in its setting and plot. Oh, an the option to switch between FPS and TPS...sweet.
Dark Void: Unique vertical cover system and I don't think Niko Bellic ever hijacked a UFO.
I Am Alive: unique concept of surviving a disaster and the other survivors who seem to be just normal people - not mutants - in a desperate situation.
