Just about any headset is gonna get uncomfortable after a couple hours. My Razer Carcharias aren't that bad though.

If its exclusively for pc and non console use than it'll open up the option for you to not use gaming headsets. Bose has the best comfort just not the best sound.

I'll recommend hd 598 sennheiser, dt770 beyer, my ultrasone dj1, and akg k701(you need an amp for that) they have BIG velour pads. They all don't have mic, so you need a seperate mic clip. Anything OVER the ear as oppose to on ear is good.

Large over ear headphones are you best bet, preferably open back. Personally I don't recommend gaming headsets.

You should give us an idea of budget, but off the top of my head some great quality comfy headphones include:

Beyer DT770, 880, 990

Sennheiser 500 and 600 series

AKG 600 and 700 series

Shure SRH 1440 and 1840

i had the same issue with a few (non-gaming) headsets that i had used until i got a logitech g35. a couple hours using it causes almost no discomfort. at around three, it gets really hot but if you wear glasses it never feels like it's causing the frames to dig into your skull.
