Honestly? There is no one single game any one person should own. Not because there aren't any great games, but because no two gamers have exactly the same tastes.
If I had to name just one game though i'd say Fallout 2. It's not a game, it's a life experience. Otherwise...
Action/Shooter: Half-Life 2
RPG: Fallout 2
Sports: Whatever the heck those people play that's "good"
RTS: Company of Heroes if your rig can handle it. This game... holy Heck... it's the most badass game i've ever played. I'm not going into detail but damn... why are you reading this and not playing it...?
MMO: CoH. I can't say WoW because it takes a certain type of person to actually stick with it long term. I say City of Heroes because it's simple, offers a little bit of everything, and because it's just fun. It's a good, well rounded MMO. Most likely wont stick with it long term, but you will keep coming back to it and have nothing but great memories.
Deus Ex
Its an awesome first-person shooter! Guns and armor and sharp sticks, oh my!
No, its an amazing RPG! Character interaction, skills, and an awesome plot!
Actually, its a hybrid of both! Its problably the most replayable single-player game in existence imo. Proof? I play it about 4 times a year ever since I got it bundled with my sound card...god, what a totally unexpected gem!

System Shock 1 was good, but System Shock 2 was the truely great title. Deus ex was the spiritual succesor of System Shock 1 by the way so you're right to presume that :)
The Chronicles of Riddick is one of the most enjoyable games I've ever played and should be in every PC gamer's collection.
It has incredibly varied gameplay, compelling story, amazing characters (especially Riddick), outstanding voice acting and graphics, gritty atmosphere and lot's of unlockable extras plus a commentary mode.
Honestly, I can't praise this game enough. It is a must-have!

YES. Im playing that at the moment and its just awesome. Great story, good acting, I havent cared this much about a game story since GTA: SA and my cursing at Sweet for making CJ get back into all that gang rubbish. Grr. Damn him! Chucked him out of the car I did. Stupid game said mission failed. Pah.
anyway, erm, GTA:SA, CoH, Call Of Duty 1, Tomb Raider 1/Anniversary, The Sims (2) - you will either love or hate, Civ series.
those are the games that made the biggest inpression of me.
