I wouldn't really consider the handheld follow up, Okamiden (82/100), as Okami 2. It's definitely a follow up (like the GTA games on handhelds), but I think the actual Okami 2 still hasn't arrived. Okamiden was a nice followup on DS, a shrunken-down Okami of sorts. It didn't feel like a proper sequel to Okami; Okamiden was smaller and different, and simply not as good and polished coming off of the glorious Okami, but Okamiden was a neat little game in it's own right. Okamiden (note it doesn't have a "2") was a nice, but flawed (too backtracking for example) handheld take on the original masterpiece Okami (92/100).
Here's what I would say, I would take an Okami sequel directed by the creator and director of Okami 1 (92/100), Hideki Kamiya.
I bought Okami day one on it's release day in 2006 simply because it was made by my boy, Kamiya. I don't follow franchises, but rather developers. He has always delivered for me.
Unfortunately, it's unlikely Capcom would work with their ex-employee Kamiya anytime soon, and if they did, I doubt they would want to waste that effort on an Okami sequel, but I would be all for it.

First of all I should clarify that Okamiden is not really a sequel to Okami...
in the sense that it's more of a successor.
Okamiden is its own franchise
so what you had on the Wii was Okami and that was its own game. We wanted to maintain the world of Okami, but create a new franchise with it so we thought the Nintendo DS was the perfect place to start with that, thus we created Okamiden to put on the DS.

I thought it was obvious to those that played both games that Okamiden wasn't quite really an Okami 2, but just in case, Capcom clarified that in fact, Okami 2 does not exist. Okamiden is spin off new franchise. I do wish more people would check both games out though, for sure.
That said, both games are good, but only the original Okami (imo) is truly an amazing game.
