For Microsoft, a generally good answer is probably the first Halo. It's genre-defining for sure, and definitely gave the Xbox a reason to exist.
But there is one game out there that could probably slay it as the greatest title Microsoft ever created.
And that's Microsoft Flight Simulator. Older than even Windows, and had no other competition to rival it. It was always a niche product, sure, but it had a charisma in the market very few cult titles have, even today. It also gave rise to the excellent Combat Flight Simulator spinoff. And lastly, I've never seen a genre stall so hard like the flight sim genre has since Microsoft pretty much left it for dead around 7 years ago.
During 2004-2007, while I probably spent 50,000 words arguing with people about the fucking lore of Halo or whatever, my friend spent most of those hours playing Flight Sim 2004/X. He's now a professional pilot, travelling the world and earning ridiculous money.
So here's to you, Microsoft Flight Simulator. May you be an eternal reminder that Microsoft can create fabulous things that aren't tied to Microsoft Office when the folks at Redmond actually put their minds to it.
Nintendo: Super Mario 64
Valve: Team Fortress 2(proof is in the pudding)
MS: Gears of War
Sony: still waiting for greatness.
Sega: Sonic 2
Valve: Half-Life 2
Sega: Sonic 3 & Knuckles
Microsoft: Halo 3
Sony: God of War 2
Nintendo: Legend of Zelda Twilight Princess (Wii)
Honorable Mentions: Super Mario 3 (NES), Uncharted 2 (PS3), Halo (Xbox), Counter-Strike Source (PC), Sonic 2 (Genesis)
*I don't fucking care who or what owns what IP rights or whatever. I fucking love this game and MS had a hand in it's being released. Bite me.
EDIT: Am I the only one or is this new font fucking horrid?

They changed the font because they found the old one caused page loading performance issues.
And here's me not noticing a single iota of difference besides the eye-bleedingly bad new font.

lol i guess we'll get used to it?
Not sure exactly what you're asking here Char, so I'm going to give two lists. One being the games that Ive enjoyed the most, and the second being those that ve held the largest impact on the industry as a whole.
Firstly, my favs:
Nintendo-Super Metroid
Sega-Panzer Dragoon Saga
Sony-The Last of Us
MS-Halo CE
Valve-Half Life 2
As to what held the largest ramifications to the industry:
Nintendo-Super Mario Brothers (VERY tempted to give this to 64)
Sega-the obvious choice...Sonic
Sony-none that come to mind really. I suppose TLoU, as it raised the bar for cinematic storytelling in gaming
MS-Halo CE
Valve-Half Life (2 was a better game, but the original was groundbreaking)
Good thread!
