So (before I play GOW3 tomorrow), its...
The metal gear super machines mission and the last couple of missions in MGS4
The Desperate Times mission in Uncharted 2
And the final levels in both KH1 and KH2 (you need plenty of stamina to finish the missions in both of those lol).
my moments have to be
retaking the white house from the russians in MW2
bank heist in grand theft auto 4
driving metal gear rex down that tunnel in MGS4
killing my first gekko in MGS4
racing in a warthog before halo blows up in halo combat evolved
Probably not the best, but it's worth mentioning.
In the original, top-down GTA there was a mission where you had to drive a truck into a police station and blow it up. It was pretty farking epic considering we had never seen anything like that in a videogame before. Not just that, I really liked the look of the original GTA and still do. The explosions in particular were awesome so it was grate seeing the building explode like that.
half life 2 the final level, when you have the super gravity gun, that was just damn awesome

assasins creed: last mission
I dunno how to really interpret what "best" denotes, but my most intense gaming moments are:
Half-Life 2, Last Level (Gravity Gun FTW!)
Halo:CE, Discovering and fleeing from the flood as well as the last level "The Maw" (EPIC)
CoD4:MW, The whole game practically
KH, Sora vs. Riku
KH2, Xemnas vs. Riku+Sora
Oblivion, destroying the drill thing
CoD4:MW2, Gulag mission, retaking DC, and the closing kill
Portal, Defying GlaDOS

I couldn't agree more with Three Leaf Clover from GTA IV,that mission was just epic!!
the mission on Novaria in Mass Effect, there is a moment where Shepard leans on a stairway and you'll have to make a decision that will affect the game and it's sequel!, all with the music (breeding grounds) in the backgrounds makes this easily the best mission i have ever played, not to mention the dialogs at the end man i love Mass EffectWings_008What are you talking about?
"All Ghilled Up" From COD4 was really sweet.
Also Snake's Tanker mission from MGS2 was REALLY nice. The best part of the game.

Half-life 2: Highway 17
Dawn of war 2: Chaos Rising; Defend the Palace
C&C Red Alert 2: soviet opening mission
Company of Heros: Opposing Fronts, Arnhem (or Hells highway if you want)
splinter cell, Double Agent (Green land mission i think it was, or the Newyork one. both excellent examples of level design and atmosphere)
Stalker: Call of Pripyat, The whole game.
EDIT Stalker:Shadow of Chernobyl,lab X-18 that was awesome!!!!!!!
