I'm gonna go for a tie between the mini-chainsaw in Dead Rising or the context-sensitive weapon for the bathroom in Conker's Bad Fur Day (you know what i mean).
lancer and torque bow in gears, gravity gun in halflife, plasminds in bioshock, clouds big-ass sword in final fantasy, i dont know the name of it, blades of athena god of war, and dawn and duskfang in shivering isles
how about the nuke missile that you can control in unreal tournament?? (sorry i cant remember what it was called anymore) it could wipe out the entire enemy team with a well guided blast :D
The gravity gun from Half Life 2, The ballers from hitman blood money or the gun that shoots shirukens and lighting from painkiller(yes I played the game)
I'd have to agree that the Gravity Gun is probably one of the most origional weapons, it's hella fun especially in the final level of Half-Life 2.
I hate the 50 cal. from COD4, wayyy underpowered. Now that I think of it, i dont think any game has shown what a 50 really does to the human body.
The Gause Rifle from Crysis... nuff said.

the chaingun out of vice city is awesome, as is the torque bow and chainsaw bayonet out of gears and the gravity gun out of hl2 is simply revoloutionary- my fav... chaingun (it has an unparalled smoothness in regard to killing people)
