If this was another long anime, I might agree with you. However, One Piece is an exception amongst long animes in the sense that it keeps improving as the series goes on. Also unlike many other long Shonen series, the plot and the characters are also very well-thought out and you will see many recurring details and characters LONG after you've forgotten about them (possibly hundreds of episodes later).
It wasn't bad before episode 45 per se, but there is a fair amount of build up that needs to be done and while its absolutely necessary for later parts of the show, it can drag on a bit and a lot of people can be turned off when the show doesn't instantly hook you. In my opinion, it's worth it to stick through those episodes because after that you get 400+ episodes and counting of pure awesomeness.

Not really. I watched about 50-60 episodes of One Piece and it dragged on and on. That soured the whole show for me for a few reasons.1) The show is literally the Family Guy of anime. A bunch of plotlines that have nothing to do with the overall story. Okay, maybe not ALL of it is random but most of it is, the other side is Luffy finding his crewmates. Luffy and friends go island to island telling the residents of said island how much they want to go to the Grand Line, and then some random douchebag comes and they fight for whatever reason. The show can literally go on and on forever with this formula and it's hella repetitive. No one likes sitting through tedium before you get to the good stuff. All those wasted episodes can be spent watching a much better anime than One Piece or just doing something else entirely.
2) There's no character development other than the random island they visit that coincidentally deals with one of the crew's past like the whole Nami thing with Arlong. Luffy is also a boring character whose only appeal is that he's just another happy-go-lucky shonen character who is able to kick ass. Most of the character's are clichés in way or another and the only slightly interesting one is Sanji.
Don't get me wrong there are some good aspects of One Piece like the humor, art-style, and music. I just don't consider it a must-watch show, more like something I'll watch when there's nothing left to do.
